Structure Therapeutics Inc. (NASDAQ: GPCR) ranks eighth because aleniglipron has emerged as one of the strongest clinical-stage oral GLP-1 candidates outside Novo Nordisk A/S (NYSE: NVO) and Eli Lilly and Company (NYSE: LLY). Aleniglipron is a once-daily, nonpeptide small-molecule GLP-1 receptor agonist. Unlike peptide-based oral semaglutide, a small-molecule medicine could potentially be manufactured at a greater scale and taken with fewer restrictions involving meals or water, though its ultimate dosing instructions will depend on clinical and regulatory findings.

In March 2026, Structure Therapeutics Inc. (NASDAQ: GPCR) reported that aleniglipron produced placebo-adjusted mean weight loss of 16.3% at the 180-milligram dose and 16% at 240 milligrams after 44 weeks in the Phase 2 ACCESS II study. The company said participants had not yet reached a weight-loss plateau. In the ongoing extension study, participants receiving 120 milligrams recorded weight loss of as much as 16.2% after 56 weeks. Structure Therapeutics Inc. (NASDAQ: GPCR) planned to use an FDA end-of-Phase 2 meeting to finalize the design of a Phase 3 program scheduled to begin during the second half of 2026.

Chief Executive Officer Raymond Stevens previously said oral small-molecule GLP-1 treatments could “meaningfully expand access and options” for patients. That is the core investment case. Structure Therapeutics Inc. (NASDAQ: GPCR) does not have to defeat every injectable treatment to build a valuable franchise. It needs to show that aleniglipron provides competitive weight loss, acceptable gastrointestinal tolerability and convenient dosing at a manufacturable scale. The risks include treatment discontinuations, high required doses and the possibility that approved pills from Novo Nordisk A/S (NYSE: NVO) and Eli Lilly and Company (NYSE: LLY) establish dominant positions before aleniglipron reaches the market.
